Preparation method | The mixture of 3-amino-2-pyrazinarboxylic acid (10g, 72 mmol) in 100 ml of anhydrous methanol and 10 ml of concentrated sulfuric acid is reacted at room temperature for 20 hours. The reaction mixture was poured into 100 ml of ice water, made alkaline with ammonium hydroxide, and extracted with dichloromethane (70 ml × 3). The combined organic extracts were washed with brine, dried with anhydrous magnesium sulfate, filtered and evaporated. The crude product was recrystallized and purified with water to obtain 5g of yellow solid methyl 3-aminopyrazine-2-carboxylic acid with a yield of 7.9g and 72%. |
